This experimental nanoparticle formulation, designated as MPDA-HK@PDA-HA, is a multifunctional nanomedicine designed for the synergistic treatment of breast cancer. It integrates chemotherapy and photothermal therapy (PTT) within a single platform. The system comprises a mesoporous polydopamine (MPDA) nanoparticle core loaded with the natural chemotherapeutic agent honokiol (HK). The surface is coated with functionalized hyaluronic acid (HA), which serves as a targeting ligand for CD44 receptors overexpressed on the surface of breast cancer cells. Upon target recognition and cellular uptake, the polydopamine component acts as a photothermal transducer, converting 808 nm near-infrared (NIR) light into localized heat to thermally ablate tumor cells. Simultaneously, the controlled release of honokiol induces apoptosis by upregulating cleaved caspase-3 and cleaved PARP. Preclinical studies have demonstrated that this nanosystem, particularly when combined with low-dose metformin, significantly inhibits tumor growth and improves therapeutic outcomes compared to monotherapies.
