Honokiol prodrug (HKLp) is a small molecule derivative of honokiol, a natural biphenolic compound extracted from the bark of *Magnolia officinalis*. It was developed to overcome the pharmacological limitations of honokiol, such as poor aqueous solubility and rapid systemic clearance, thereby enhancing its pharmacokinetic profile and circulation time. HKLp is being investigated for its radioprotective properties, specifically its ability to mitigate radiation-induced normal tissue damage. Its mechanism of action involves the activation of Sirtuin 3 (SIRT3), which helps restore mitochondrial respiratory function and reduces radiation-induced immunogenicity and oxidative stress. Preclinical studies have demonstrated that HKLp can alleviate systolic dysfunction and prevent restrictive cardiac remodeling in models of thoracic irradiation, suggesting potential therapeutic utility in radiation-induced heart disease (RIHD).
