HotSpot Therapeutics is developing a first-in-class small molecule inhibitor of Interferon Regulatory Factor 5 (IRF5) for the treatment of autoimmune diseases, including systemic lupus erythematosus (SLE), Sjögren's syndrome, rheumatoid arthritis (RA), systemic sclerosis (SSc), and myositis. IRF5 is a transcription factor that acts as a master regulator of innate immunity; its dysregulation is linked to the production of pro-inflammatory cytokines and type I interferons. Discovered via the proprietary Smart Allostery™ platform, the inhibitor targets a "natural hotspot"—a previously unknown allosteric pocket critical for the protein's endogenous regulation. Preclinical data demonstrate that the compound provides dose-dependent inhibition of IRF5 phosphorylation and nuclear translocation. In December 2022, AbbVie entered into an exclusive worldwide collaboration and option to license agreement for the program.
