Overview

House dust mite allergen Der p 1 is a major glycoprotein allergen derived from the mid-gut and fecal pellets of the mite *Dermatophagoides pteronyssinus*. It is a papain-like cysteine protease and is present in both native and recombinant forms. Der p 1 exhibits structural homology and cross-reactivity with similar allergens such as Der f 1. Exposure to Der p 1 occurs via inhalation of mite excrement, and it can trigger robust IgE responses in sensitized individuals, causing allergic diseases such as asthma and allergic rhinitis. Mechanistically, Der p 1 disrupts airway epithelial barrier function by protease activity, leading to epithelial cell desquamation and facilitating allergen penetration. It induces the release of proinflammatory cytokines (e.g., IL-6 and IL-8) through proteinase-activated receptors. It also modulates immune responses by cleavage of immune cell surface receptors (CD23, CD25), promoting IgE production, and inactivates innate lung defense proteins such as elastase inhibitors. Der p 1 is routinely measured to diagnose genuine house dust mite allergy and guide allergen immunotherapy[1][2][3][5][6].
