Drug intelligence / Profile preview
House dust mite allergoid is a subcutaneous allergen immunotherapy (SCIT) developed by Allergopharma for the treatment of house dust mite-induced allergic rhinitis, allergic rhinoconjunctivitis, and allergic asthma. It consists of chemically modified allergen extracts (allergoids) from the house dust mites *Dermatophagoides pteronyssinus* and *Dermatophagoides farinae*, which are cross-linked with formaldehyde and glutaraldehyde and adsorbed onto aluminum hydroxide as a depot adjuvant. This chemical modification reduces the IgE-binding capacity of the allergens, thereby minimizing the risk of IgE-mediated systemic allergic reactions (anaphylaxis) while preserving the T-cell epitopes necessary to induce immunological tolerance. The therapy works by promoting a shift from a Th2-dominated allergic response to a Th1/Treg-mediated tolerant state, inducing the production of allergen-specific IgG4 blocking antibodies and regulatory T cells.
