Overview

The Hox-PBX binding inhibitor is a preclinical-stage small molecule developed by HOX Therapeutics that targets the protein-protein interaction (PPI) between HOX transcription factors and their essential cofactor, PBX (Pre-B-cell leukemia homeobox). HOX genes, which are critical for embryonic development, are frequently dysregulated in various malignancies, including glioblastoma, breast cancer, and acute myeloid leukemia, where they drive oncogenic signaling pathways that promote cell survival and proliferation. By competitively inhibiting the binding of HOX proteins to the PBX cofactor, this small molecule prevents the formation of functional transcription factor complexes, thereby suppressing the expression of genes involved in tumor maintenance and inducing apoptosis. This small molecule program is distinct from the company's peptide-based candidates (such as HTL-001) and aims to provide a targeted therapeutic approach for tumors characterized by HOX gene dysregulation.
