hP1A8 is an investigational recombinant polypeptide and first-in-class peptide checkpoint inhibitor developed by OX2 Therapeutics. It acts as a ligand for the CD200 activation receptor (CD200AR), a target within the CD200 immune checkpoint pathway. Unlike traditional checkpoint inhibitors that block inhibitory signals, hP1A8 is designed to activate the CD200AR to stimulate an immune response and counteract the immunosuppressive environment typically found in tumors like glioblastoma. The drug is intended to enhance anti-tumor T-cell activity and may also modulate the suppressive effects of other checkpoints, including PD-1/PD-L1 and CTLA-4. It is currently being evaluated in Phase I clinical trials as an adjuvant therapy in combination with the GBM6-AD tumor lysate vaccine and imiquimod for the treatment of recurrent glioblastoma.
