Overview

HPA-12 is a **small molecule ceramide analog** that acts as a potent and selective **inhibitor of ceramide transfer protein (CERT)**. CERT mediates non-vesicular transport of ceramide from the endoplasmic reticulum to the Golgi apparatus for sphingomyelin synthesis. By competitively inhibiting CERT, HPA-12 disrupts intracellular ceramide trafficking and ceramide metabolism. It has primarily been used as a research tool to study ceramide biology, sphingolipid metabolism, and related cellular processes including apoptosis, inflammation, and neurodegenerative diseases. HPA-12 crosses the blood-brain barrier and is being explored in models of Alzheimer’s disease and other disorders linked to perturbed ceramide metabolism[1][2][4].
