Overview

hPAM4-SN-38 (IMMU-134) is an antibody-drug conjugate (ADC) developed by Immunomedics for the treatment of MUC1-expressing cancers, particularly pancreatic cancer. It consists of the humanized monoclonal antibody hPAM4 (clivatuzumab), which targets a specific epitope of Mucin 1 (MUC1), conjugated to the cytotoxic payload SN-38 (the active metabolite of irinotecan) via a CL2A linker. The mechanism of action involves the binding of the antibody to MUC1 on the surface of cancer cells, followed by internalization and release of SN-38. SN-38 then inhibits DNA Topoisomerase I, leading to double-strand DNA breaks and apoptosis. Preclinical studies have demonstrated efficacy in pancreatic cancer xenograft models.
