Overview

HPG-MTX is a nanoparticle-based formulation in which methotrexate (MTX), a well-known antimetabolite and immunosuppressant, is chemically conjugated to hyperbranched polyglycerol (HPG). This drug is designed to improve methotrexate delivery, enhance pharmacokinetics, and reduce toxicity. The mechanism of action derives from methotrexate's properties: it inhibits dihydrofolate reductase, blocking tetrahydrofolate synthesis, which is crucial for DNA and RNA synthesis, thus exerting cytotoxic effects in cancer and immunomodulatory effects in autoimmune disease. Conjugation to HPG exploits the physicochemical properties of nanoparticles to increase bioavailability and stability while aiming to mitigate MTX’s adverse effects seen with free drug formulations. The primary indication for HPG-MTX is presumed to be investigational uses in cancer or autoimmune diseases, leveraging the established mechanism of action of methotrexate in a novel delivery system[1][2].
