HPG-MTX is a nanoparticle-based formulation in which methotrexate (MTX), a well-known antimetabolite and immunosuppressant, is chemically conjugated to hyperbranched polyglycerol (HPG). This drug is designed to improve methotrexate delivery, enhance pharmacokinetics, and reduce toxicity. The mechanism of action derives from methotrexate's properties: it inhibits dihydrofolate reductase, blocking tetrahydrofolate synthesis, which is crucial for DNA and RNA synthesis, thus exerting cytotoxic effects in cancer and immunomodulatory effects in autoimmune disease.
