HPG7233 is a highly selective, liver-targeted small molecule agonist of thyroid hormone receptor beta (THR-β). Developed by Hepagene Therapeutics, it is designed to modulate serum and liver lipid metabolism. Preclinical studies have demonstrated that HPG7233 has high selectivity for THR-β over THR-α, with a favorable pharmacokinetic profile characterized by liver enrichment and low extrahepatic distribution. The drug significantly reduces hepatic triglycerides and serum LDL cholesterol in animal models of metabolic disease. It has shown robust efficacy in reducing steatosis and improving markers of nonalcoholic steatohepatitis (NASH), as well as demonstrating synergistic effects when combined with the FXR agonist HPG1860. Toxicology studies indicate good safety and tolerability without adverse findings related to THR-α or gastrointestinal toxicity[1][4][6].
