Overview

HPG7233 + HPG1860 is a combination therapy being developed by Hepagene Therapeutics for the treatment of metabolic dysfunction-associated steatohepatitis (MASH). The combination consists of two liver-selective small molecules: HPG7233, a highly selective thyroid hormone receptor beta (THR-β) agonist, and HPG1860, a next-generation non-bile acid farnesoid X receptor (FXR) agonist. HPG7233 targets THR-β to reduce hepatic fat accumulation and improve lipid profiles, while HPG1860 targets FXR to regulate bile acid homeostasis and modulate inflammatory pathways. Preclinical studies have demonstrated significant synergistic effects between these two mechanisms, suggesting enhanced efficacy in reducing liver fat and inflammation compared to monotherapy. While HPG1860 has completed Phase 2a clinical trials, the combination is currently in early-stage development following the FDA IND clearance of HPG7233 in late 2023.
