HPi1 is a small molecule antimicrobial agent developed by Arietis Pharma specifically for the treatment of Helicobacter pylori infections. Identified through a high-throughput screen, HPi1 demonstrates potent and highly selective activity against H. pylori, with an IC50 of 0.24 μM and an MIC of 0.08-0.16 μg/mL. In preclinical studies using a mouse model of infection, oral administration of HPi1 at 25 mg/kg/day for three days effectively reduced bacterial colony counts to below the limit of detection. While the exact biological target remains under investigation, its high degree of selectivity suggests it may inhibit a target essential for H. pylori that is absent or poorly conserved in other bacterial species, potentially reducing impact on the commensal microbiota.
