Overview

HPN424 is a tri-specific T cell engager developed using Harpoon Therapeutics' proprietary TriTAC (Tri-specific T cell Activating Construct) platform. It is designed to redirect a patient’s own immune cells to kill tumor cells expressing prostate-specific membrane antigen (PSMA), primarily in prostate cancer. The molecule contains three binding domains: - An anti-PSMA domain for tumor cell engagement, - An anti-albumin domain for half-life extension, - An anti-CD3 domain for T cell engagement. HPN424 acts by bringing cytotoxic T lymphocytes into close proximity with PSMA-expressing cancer cells, thereby stimulating targeted immune-mediated killing of these tumor cells. It was optimized for small size and increased stability compared to other bispecific platforms[2][5][7]. The drug was administered as a weekly intravenous infusion in clinical trials.
