Overview

HPN536 is a trispecific T-cell activating construct (TriTAC) designed for cancer immunotherapy. It is a recombinant protein-based drug that simultaneously binds to mesothelin (MSLN) on tumor cells, CD3ε on T cells, and serum albumin. By engaging both the tumor antigen mesothelin and the CD3 receptor on T cells, HPN536 redirects and activates T cells to kill mesothelin-expressing cancer cells. The albumin-binding domain extends its plasma half-life. Developed initially by Harpoon Therapeutics, it was later acquired by Merck & Co. The drug has been investigated primarily for advanced cancers associated with mesothelin expression, including ovarian cancer, pancreatic cancer, lung cancer, and mesothelioma[1][2][3][4][5][6].
