Overview

HPP-3033 is a small molecule drug developed by vTv Therapeutics. It acts as an inhibitor of both **BACH1 (BTB domain and CNC homolog 1)** and **Nrf2 (Nuclear factor erythroid 2-related factor 2)**. Its mechanism leverages the inhibition of BACH1 to derepress antioxidant and anti-inflammatory pathways, particularly by upregulating genes involved in heme degradation and redox regulation such as Hmox1, Gclc, Gclm, Nqo1, and other ARE-dependent and non-ARE-dependent targets. The drug is primarily being developed for **nonalcoholic steatohepatitis (NASH)** and is being investigated for its potential effects on digestive system disorders. There are also indications that it may have neuroprotective effects in models of neurodegeneration through similar gene regulatory mechanisms, but its primary development is in NASH[1][2][6][7].
