HPP854 is an orally active small molecule inhibitor of beta-site amyloid precursor protein cleaving enzyme 1 (BACE1), also known as beta-secretase. It was developed for the treatment of Alzheimer's disease and mild cognitive impairment. By inhibiting BACE1, HPP854 aimed to reduce the production of amyloid-beta peptides implicated in the pathogenesis of Alzheimer's disease. The drug was originally developed by High Point Pharmaceuticals, a spin-off from TransTech Pharma, with vTv Therapeutics also associated with its development. Clinical development reached phase 1 but was discontinued after termination of early trials due to lack of efficacy or other strategic reasons[2][3][4][5].
