HPV-16 E7SH is a therapeutic DNA vaccine candidate designed for the treatment of cervical cancer and other diseases associated with high-risk human papillomavirus type 16 (HPV-16). The vaccine utilizes a naked DNA plasmid encoding an artificial, "shuffled" version of the HPV-16 E7 oncogene. This shuffling process rearranges the gene sequence to preserve all known cytotoxic T-lymphocyte (CTL) epitopes while completely eliminating the transforming (oncogenic) properties of the wild-type E7 protein, thereby enhancing the safety profile for human clinical use. The vaccine aims to induce both cellular and humoral immune responses, specifically priming antigen-specific T cells to recognize and eliminate HPV-infected or malignant cells. Preclinical studies have demonstrated significant tumor protection and regression in murine models, and the construct has shown immunogenicity in human T cells in vitro.
