TollereBio's HPV adjuvant program is a preclinical-stage vaccine strategy that utilizes Bacterial Enzymatic Combinatorial Chemistry (BECC)-derived TLR4 agonists, specifically BECC470 and BECC438, in combination with an RG1-VLP (virus-like particle) HPV vaccine and the aluminum hydroxide adjuvant Alhydrogel. This approach leverages synthetic lipid A mimetics to activate Toll-like receptor 4 (TLR4), thereby enhancing and broadening the immune response against Human papillomavirus (HPV) infection. Preclinical mouse studies have demonstrated that this adjuvant system can induce strong T cell responses, increase memory B cell frequency, and generate cross-neutralizing antibodies against multiple HPV strains, including HPV16, HPV18, and HPV39. Furthermore, the BECC/Alhydrogel combination allows for significant antigen dose-sparing (up to 75% reduction) and may accelerate immune responses, potentially reducing the number of required vaccine doses.
