Overview

HPV16 RG1 VLP is a next-generation prophylactic human papillomavirus (HPV) vaccine candidate. It consists of virus-like particles (VLPs) formed by the major capsid protein L1 of HPV type 16, into which the highly conserved "RG1" epitope from the minor capsid protein L2 (amino acids 17–36) has been genetically inserted into the immunogenic DE-surface loop. This design enables repetitive and highly immunogenic display of the L2 epitope on the surface of assembled VLPs[5][7]. The mechanism of action involves induction of neutralizing antibodies against both HPV16 and a broad range of other high-risk and low-risk mucosal as well as some cutaneous HPVs by targeting conserved regions in L2[6][5][7]. Preclinical studies have shown that vaccination with this construct induces robust B cell memory, cytotoxic T lymphocyte responses, and long-lasting cross-protection against multiple clinically relevant high-risk mucosal HPVs responsible for most cervical cancers[5][7]. The vaccine is being developed to address limitations in current licensed vaccines by providing broader protection across diverse HPV types not covered by existing products[2].
