HPV16 TCR-NK is an investigational adoptive cell therapy consisting of natural killer (NK) cells engineered to express a T-cell receptor (TCR) specific for Human Papillomavirus type 16 (HPV16) antigens, most commonly the E7 oncoprotein. Unlike traditional CAR-NK cells that recognize surface antigens, TCR-NK cells are designed to recognize intracellular viral peptides presented by Major Histocompatibility Complex (MHC) molecules, such as HLA-A*02:01. This approach combines the innate, HLA-independent anti-tumor activity of NK cells with the antigen-specific, HLA-restricted targeting capabilities of a TCR. The therapy is primarily being developed for HPV16-positive solid tumors, including cervical cancer and oropharyngeal squamous cell carcinoma. Clinical development is currently led by academic institutions, notably Sun Yat-sen University, which is conducting Phase 1 trials to evaluate the safety and preliminary efficacy of these engineered cells.
