HRA00129-C004 is a novel antibody-drug conjugate (ADC) targeting the c-Met receptor tyrosine kinase (MET). Developed by Hengrui Pharmaceuticals, it consists of a humanized anti-c-Met monoclonal antibody conjugated to a potent topoisomerase I inhibitor payload through a cleavable linker. The ADC is designed to bind to c-Met on the surface of tumor cells, undergo internalization into lysosomes, and release its cytotoxic payload. This release leads to DNA damage and apoptosis in c-Met-expressing cancer cells, while also exhibiting a bystander effect on neighboring cells. Preclinical studies have shown strong antitumor activity in various xenograft models and a favorable safety profile. It is currently being evaluated in Phase I clinical trials for patients with advanced solid tumors.
