HRA00130-C004

HRA00130-C004 is a novel antibody-drug conjugate (ADC) targeting Delta-like ligand 3 (DLL3), a protein highly expressed in small cell lung cancer (SCLC) and other high-grade neuroendocrine tumors. Developed by Shanghai Hengrui Pharmaceutical, the drug consists of a humanized IgG1 antibody conjugated to a topoisomerase I inhibitor payload, DXh, via a cleavable linker. HRA00130-C004 is designed with a high drug-to-antibody ratio (DAR) and exhibits a potent bystander effect, allowing it to kill neighboring DLL3-negative cells. Preclinical studies have demonstrated significant anti-tumor activity in both DLL3-high and DLL3-low xenograft models, along with a favorable safety and pharmacokinetic profile in animal models.
