Overview

HRS-2329 is an orally bioavailable small molecule inhibitor of Son of Sevenless homolog 1 (SOS1), currently under development by Jiangsu HengRui Medicine. SOS1 acts as a guanine nucleotide exchange factor (GEF) that facilitates the activation of RAS proteins (KRAS, NRAS, HRAS) by promoting the exchange of bound GDP for GTP. By inhibiting SOS1, HRS-2329 prevents the formation of the active GTP-bound state of RAS, thereby suppressing downstream oncogenic signaling pathways such as the MAPK/ERK pathway. This mechanism is particularly relevant for treating tumors driven by various RAS mutations or amplifications, which are common drivers in many solid cancers. Clinical investigation is focused on its safety, tolerability, and efficacy in patients with advanced solid tumors harboring these specific genetic alterations.
