HRS-2604 is a small molecule inhibitor of Son of Sevenless homolog 1 (SOS1) developed by Tuojie Biotech, a subsidiary of Jiangsu Hengrui Pharmaceuticals. SOS1 is a guanine nucleotide exchange factor (GEF) that plays a critical role in the activation of RAS proteins, including KRAS, by promoting the exchange of GDP for GTP. By targeting SOS1, HRS-2604 aims to disrupt the activation of the MAPK signaling pathway, which is frequently dysregulated in various cancers, particularly those harboring KRAS mutations. The drug is primarily being investigated for the treatment of advanced solid tumors, either as a monotherapy or in combination with other targeted agents to overcome resistance mechanisms associated with KRAS-driven oncogenesis.
