Overview

**HRS-4642 + cetuximab** is a combination investigational therapy comprised of HRS-4642, a selective long-acting KRAS G12D inhibitor formulated as a liposomal injection, and cetuximab, an epidermal growth factor receptor (EGFR) monoclonal antibody. HRS-4642 inhibits mutant KRAS G12D, suppressing downstream MEK/ERK phosphorylation to block tumor cell proliferation. Cetuximab antagonizes EGFR-mediated signaling, counteracting adaptive feedback resistance commonly triggered by KRAS inhibition. The combination aims to improve efficacy in advanced solid tumors, especially pancreatic ductal adenocarcinoma (PDAC) with KRAS G12D mutations that are refractory to standard therapies.[1][3][5]
