Overview

HRS-4729 is a synthetic peptide drug under development as a next-generation incretin-based therapy for metabolic diseases. It acts as an agonist at three key receptors involved in glucose and energy homeostasis: the glucagon-like peptide 1 receptor (GLP-1R), the gastric inhibitory polypeptide receptor (GIPR), and the glucagon receptor (GCGR). By targeting these pathways, HRS-4729 aims to promote weight loss and improve glycemic control. The drug is being developed primarily for obesity and type 2 diabetes mellitus. It is currently in Phase 1 clinical trials for obesity in China, with preclinical development ongoing for type 2 diabetes mellitus[1][6][7][9]. The originator and main developer is Jiangsu Hengrui Pharmaceuticals, with licensing activities involving Kailera Therapeutics[4][6][8].
