HRS-5765 is an orally bioavailable small molecule soluble guanylate cyclase (sGC) stimulator being developed by Chengdu Suncadia Medicine, a subsidiary of Jiangsu Hengrui Pharmaceuticals, for the treatment of heart failure. The drug works by binding to and stimulating the sGC enzyme, both independently of and synergistically with nitric oxide (NO). This stimulation increases the intracellular concentration of cyclic guanosine monophosphate (cGMP), a key secondary messenger that mediates vasodilation, inhibits platelet aggregation, and reduces cardiac hypertrophy and fibrosis. By restoring the NO-sGC-cGMP signaling pathway, which is often impaired in heart failure patients, HRS-5765 aims to improve hemodynamic function and clinical outcomes. It is currently in Phase I clinical development, with studies focusing on its safety, tolerability, and pharmacokinetics in healthy volunteers, as well as its potential interaction with CYP3A4 enzymes and the effect of food on its absorption.
