HRS-7058 + BP102 is an investigational combination therapy developed by Shandong Suncadia Medicine, a subsidiary of Jiangsu Hengrui Pharmaceuticals, for the treatment of advanced malignant solid tumors. HRS-7058 is a potent and selective small molecule inhibitor of Extracellular Signal-Regulated Kinase 1 and 2 (ERK1/2), which are terminal kinases in the RAS-RAF-MEK-ERK (MAPK) signaling pathway. Inhibition of ERK1/2 is intended to block tumor cell proliferation and survival in cancers where this pathway is constitutively active. BP102 is a liposomal formulation of irinotecan hydrochloride, a topoisomerase I inhibitor. The liposomal delivery system (nanoparticle) is designed to enhance the pharmacokinetic profile of irinotecan, increasing its intratumoral concentration and extending its therapeutic effect by stabilizing the cleavable complex between DNA and topoisomerase I, leading to double-strand DNA breaks and apoptosis. This combination is currently being evaluated in Phase II clinical trials to assess its safety, tolerability, and efficacy in patients with unresectable locally advanced or metastatic solid tumors.
