Overview

HRS-8364 is an orally bioavailable small molecule inhibitor of Son of Sevenless homolog 1 (SOS1), developed by Jiangsu HengRui Medicine. SOS1 serves as a guanine nucleotide exchange factor (GEF) that facilitates the activation of RAS proteins by promoting the exchange of bound GDP for GTP. By inhibiting SOS1, HRS-8364 prevents the activation of various KRAS mutants (including G12D, G12V, and G13D) as well as wild-type KRAS, thereby blocking the downstream MAPK/ERK signaling pathway that drives tumor growth and survival. This approach is designed to provide a broader therapeutic window and overcome resistance mechanisms associated with direct KRAS inhibitors. HRS-8364 is currently being investigated in Phase I/II clinical trials for the treatment of patients with advanced solid tumors.
