Overview

HRS-8562 is an orally bioavailable small molecule inhibitor of the Bromodomain and Extra-Terminal motif (BET) proteins, including BRD2, BRD3, BRD4, and BRDT. Developed by Tuojie Biotech, a subsidiary of Jiangsu Hengrui Pharmaceuticals, it is primarily being investigated for the treatment of myelofibrosis and other hematologic malignancies. BET inhibitors work by competitively binding to the acetyl-lysine recognition pockets (bromodomains) of BET proteins, which prevents their interaction with acetylated histones on chromatin. This disruption inhibits the transcription of key oncogenes and pro-inflammatory cytokines, such as MYC and various interleukins, which are critical drivers of the bone marrow fibrosis and systemic inflammation characteristic of myelofibrosis.
