Overview

HRS5685 is an oral small molecule autophagy inducer currently in Phase 1 clinical development. Developed by Retro Biosciences through a licensing agreement with Jiangsu Hengrui Pharmaceuticals, the drug is the lead candidate in Retro's autophagy program, which aims to treat age-related diseases and extend healthy human lifespan. HRS5685 acts as a potent activator of the AMP-activated protein kinase (AMPK) pathway. By stimulating AMPK, the drug promotes cellular homeostasis through the induction of autophagy—a process where cells degrade and recycle damaged components—while simultaneously inhibiting energy-consuming anabolic processes. The compound is being evaluated in healthy subjects to establish safety, tolerability, and pharmacokinetics before proceeding to trials for specific age-related indications.
