hRS7 ATAC (also known as HDP-103) is a second-generation amanitin-based antibody-drug conjugate (ATAC) targeting Trophoblast cell surface antigen 2 (TROP2). It comprises the humanized RS7 antibody (hRS7), which is the same antibody used in the approved ADC sacituzumab govitecan, conjugated to the potent payload alpha-amanitin. Amanitin is a bicyclic peptide derived from the *Amanita phalloides* mushroom that acts as a highly specific inhibitor of RNA polymerase II, leading to the inhibition of transcription and subsequent apoptosis in both proliferating and resting cells. Developed by Heidelberg Pharma, hRS7 ATAC is designed to address TROP2-expressing malignancies, particularly those that are refractory to standard therapies, such as pancreatic ductal adenocarcinoma and triple-negative breast cancer (TNBC). Preclinical data indicate that hRS7 ATAC exhibits high cytotoxicity, efficient internalization, and superior antitumor efficacy in xenograft models compared to first-generation TROP2 ADCs.
