HS-10365 is an orally bioavailable, highly potent, and selective small molecule inhibitor of the proto-oncogene receptor tyrosine kinase rearranged during transfection (RET). Developed by Jiangsu Hansoh Pharmaceutical Group, it selectively binds to and inhibits RET activity, blocking downstream signaling pathways involved in tumor cell proliferation and survival. HS-10365 has demonstrated promising antitumor activity in patients with RET gene fusion-positive non-small cell lung cancer (NSCLC) and other advanced solid tumors harboring RET alterations. Clinical trials have shown a manageable safety profile and favorable pharmacokinetic properties. The drug is being evaluated in Phase 1/2 clinical studies for advanced or metastatic malignant solid tumors, including NSCLC, medullary thyroid carcinoma (MTC), radioiodine-refractory thyroid cancer, neuroblastoma, and other cancers with RET gene abnormalities.
