HS-10381 is an orally administered, small molecule inhibitor targeting Src homology region 2-containing protein tyrosine phosphatase (SHP2), also known as protein tyrosine phosphatase non-receptor type 11. By binding to and inhibiting SHP2, HS-10381 disrupts SHP2-mediated signaling pathways such as the MAPK pathway, which are implicated in tumor cell proliferation and survival. The drug is being developed primarily for the treatment of advanced solid tumors that have not responded to standard therapies. Its mechanism of action may also enhance immune responses against tumors by increasing MHC class I expression on cancer cells. The developer and manufacturer is Jiangsu Hansoh Pharmaceutical. As of early 2025, it is in Phase I clinical trials in China for patients with advanced solid tumors.
