Overview

HS-10383 is a highly selective, orally available small molecule antagonist of the purinergic P2X3 receptor. It is being developed by Jiangsu Hansoh Pharmaceutical for the treatment of refractory or unexplained chronic cough (RUCC). The P2X3 receptor plays a key role in the cough reflex pathway, and antagonists targeting this receptor have shown promise in reducing cough frequency. HS-10383 distinguishes itself by effectively reducing cough with minimal impact on taste, which may improve patient adherence compared to less selective agents. Clinical studies indicate that it is generally well tolerated, with most adverse events being mild to moderate and no significant taste-related side effects reported. The drug has advanced through Phase 1 trials and entered Phase 2 development for RUCC[1][4][5][8].
