Overview

HS-20093 + cetuximab is a combination of two anticancer agents: HS-20093, a B7-H3-targeted antibody-drug conjugate (ADC), and cetuximab, an epidermal growth factor receptor (EGFR)-targeting monoclonal antibody. - **HS-20093** consists of a fully humanized anti-B7-H3 monoclonal antibody covalently linked to a topoisomerase inhibitor (TOPOi) payload. Upon binding to B7-H3, which is overexpressed on many solid tumors, HS-20093 is internalized and releases its cytotoxic payload, resulting in tumor cell death. It is primarily in development for extensive-stage small cell lung cancer (ES-SCLC), sarcoma, head and neck cancers, and esophageal squamous cell carcinoma[2][7][3][5]. - **Cetuximab** is a recombinant, chimeric IgG1 monoclonal antibody that binds to the extracellular domain of the epidermal growth factor receptor (EGFR), blocking ligand-induced activation and subsequent downstream signaling. Cetuximab is approved for the treatment of colorectal cancer and head and neck squamous cell carcinoma. - The combination aims to target tumor cells via dual mechanisms: B7-H3 inhibition/directed cytotoxicity (HS-20093) and EGFR blockade (cetuximab)[1][2]. - The combination is currently being explored in preclinical or early clinical studies for the treatment of solid tumors.
