Overview

HS024 is a synthetic cyclic peptide and a highly potent and selective antagonist of the melanocortin-4 receptor (**MC4 receptor**). It shows a *Ki* of 0.29 nM for MC4, with significantly lower affinities for MC5, MC3, and MC1 receptors, making it about 20-fold more selective for MC4. The peptide contains a 29-membered ring structure and includes modified amino acid residues. In preclinical studies, HS024 potently antagonizes the effects of alpha-MSH (alpha-melanocyte-stimulating hormone) on MC4 receptor signaling, thereby increasing food intake in rodents. It is primarily used as a research tool to study energy homeostasis, appetite regulation, and melanocortin receptor pharmacology[1][3][5][7].
