Overview

**HSA-ATO** is an experimental human serum albumin-conjugated arsenic trioxide drug-delivery construct designed to improve delivery of arsenic trioxide to solid tumors. In pancreatic cancer models, albumin conjugation increased tumor penetration of arsenic trioxide, while efficacy and cellular uptake were substantially greater in tumors with wildtype rather than mutant **MUC4**. The reported mechanism associates HSA-ATO uptake with activation of alkaline phosphatase-associated transcytosis mediated by ALPL in MUC4-wildtype pancreatic cancer cells.
