Overview

HSB-3215 is a bispecific antibody (BsAb) targeting the extracellular domains of HER2 and HER3, both of which belong to the ERBB receptor tyrosine kinase family and are validated targets in oncology[1][3][5][7][9]. It is designed as a humanized immunoglobulin with two antigen recognition arms that bind to both overlapping and unique conformational epitopes on HER2, as well as novel epitopes on HER3[1][3][5][7]. This allows for ligand-blocking and potentially complementary fit to HER3, influencing both ligand-dependent and ligand-independent signaling pathways implicated in tumor cell growth. HSB-3215 is built on Hillstream's "Knob" biologics platform known as Picobody™ technology (via collaboration with OmniAb and Minotaur Therapeutics), offering a framework to target previously inaccessible regions of HER2/HER3 heterodimer complexes[1][3]. The aim is to develop improved immunotherapeutic strategies for several solid tumors, especially those resistant to existing anti-HER2 therapies[1][3][5][7][9]. Preclinical studies focus on oncology indications including breast, lung, gynecological, endocrinological, and CNS tumors. HSB-3215's backbone is also being evaluated for potential adaptation into antibody-drug conjugates (ADCs)[5][7].
