Overview

HSK31679 is a highly selective small molecule agonist of the thyroid hormone receptor beta (THR-β), developed by Haisco Pharmaceutical Group. It is designed for oral administration and primarily targets metabolic liver diseases, including non-alcoholic steatohepatitis (NASH), non-alcoholic fatty liver disease (NAFLD), and hypercholesterolemia. The drug demonstrates higher potency and selectivity for THR-β over THR-α, aiming to maximize hepatic effects while minimizing adverse cardiac and bone effects associated with THR-α activation[5][6]. In preclinical studies, HSK31679 outperformed MGL-3196 (resmetirom) in ameliorating diet-induced steatohepatitis in mice with intact gut microbiota but not in germ-free mice, highlighting the importance of gut microbial interactions—specifically modulation of Bacteroides thetaiotaomicron and its glucosylceramide synthase activity—in its mechanism[1][4][7]. Clinical trials have shown that 12 weeks of treatment significantly reduces liver fat content in patients with NAFLD or NASH[3]. The drug is currently undergoing phase 2 clinical trials for these indications.
