HSK47977 is an orally bioavailable B-cell lymphoma 6 (BCL6) protein degrader developed by Haisco Pharmaceutical Group. It is designed as a proteolysis-targeting chimera (PROTAC) that utilizes a novel cereblon (CRBN) E3 ligase ligand to induce the ubiquitination and subsequent proteasomal degradation of the BCL6 protein. BCL6 is a transcriptional repressor and a key oncogenic driver in various B-cell malignancies, particularly diffuse large B-cell lymphoma (DLBCL). By degrading the BCL6 protein rather than simply inhibiting its activity, HSK47977 aims to overcome the limitations of traditional small-molecule inhibitors that may fail to disrupt the protein's scaffolding functions. The drug is currently being evaluated in Phase I clinical trials for the treatment of patients with relapsed or refractory non-Hodgkin lymphoma.
