Overview

HSK55879 is an orally bioavailable, highly selective small molecule inhibitor of Tyrosine Kinase 2 (TYK2), developed by Haisco Pharmaceutical Group. TYK2 is a member of the Janus kinase (JAK) family that mediates signaling for cytokines such as interleukin-23 (IL-23), interleukin-12 (IL-12), and Type I interferons (IFN), which are critical in the pathogenesis of various autoimmune and inflammatory diseases. By selectively targeting the pseudokinase (JH2) domain of TYK2 rather than the conserved catalytic (JH1) domain shared by JAK1, JAK2, and JAK3, HSK55879 aims to provide therapeutic efficacy in conditions like psoriasis and inflammatory bowel disease while minimizing off-target safety concerns associated with broader JAK inhibition, such as hematological abnormalities or lipid elevations. It is currently undergoing Phase I clinical evaluation to assess its safety, tolerability, and pharmacokinetics in healthy subjects.
