Overview

HSK56630 is an orally bioavailable, highly selective small-molecule allosteric inhibitor of Tyrosine Kinase 2 (TYK2), developed by Haisco Pharmaceutical Group. TYK2 is a member of the Janus kinase (JAK) family that mediates the signaling of key pro-inflammatory cytokines, including interleukin-23 (IL-23), interleukin-12 (IL-12), and Type I interferons (IFN). By selectively targeting the pseudokinase (JH2) domain of TYK2 rather than the conserved catalytic domain, HSK56630 achieves high selectivity over other JAK family members (JAK1, JAK2, and JAK3), potentially reducing the risk of off-target effects such as hematological abnormalities and lipid elevations. The drug is being developed for the treatment of various autoimmune and inflammatory disorders, including psoriasis and atopic dermatitis. As of early 2024, HSK56630 is in Phase 1 clinical development to evaluate its safety, tolerability, and pharmacokinetics in healthy participants.
