The **hsp110-gp100 complex** is a recombinant chaperone vaccine consisting of the large heat shock protein Hsp110 complexed with the melanoma antigen gp100. It is designed to function as a cancer vaccine by leveraging Hsp110's extraordinary protein-holding capacity to deliver gp100, a tumor-associated antigen, to the immune system for robust antigen cross-presentation by dendritic cells. This stimulates a tumor-specific CD8+ cytotoxic T cell response and can also promote humoral (antibody) immunity. The vaccine is synthetic/recombinant, allowing for uniform, scalable manufacture and stability, advantages over autologous tumor-derived HSP vaccines. Preclinical studies showed antitumor efficacy and immunostimulation in murine melanoma. A Phase Ib study in humans with advanced melanoma demonstrated minimal toxicity and measurable clinical and immune responses, especially at lower doses. Its mechanism is to enhance immune recognition of gp100-expressing melanoma cells. It was developed primarily for malignant melanoma but could be used as a platform for other tumor antigens[1][3][2].
