HSP70-derived peptide (specifically the HSP70 407-415 sequence, IVLNVLRLI) is an investigational cancer vaccine designed to elicit a targeted immune response against tumors overexpressing Heat Shock Protein 70 (HSP70). HSP70 is a molecular chaperone that is frequently upregulated in various malignancies, including pancreatic, colorectal, and gastric cancers, where it plays a critical role in protein folding, stabilization, and protection against apoptosis under cellular stress. The vaccine works by presenting the 9-mer peptide in the context of the HLA-A*2402 MHC class I molecule, thereby inducing the expansion of specific cytotoxic T lymphocytes (CTLs) that recognize and lyse HSP70-positive cancer cells. Developed primarily by researchers at Sapporo Medical University, this peptide has been evaluated in multiple Phase 1 and Phase 2 clinical trials across a broad range of solid tumors, often administered in combination with adjuvants like incomplete Freund's adjuvant (IFA).
