The HSP90-PI3K conjugate is a preclinical-stage small molecule drug conjugate (SMDC) being developed by SciClone Pharmaceuticals under an exclusive license from Tarveda Therapeutics for the Greater China region. Built on Tarveda's Pentarin® miniature drug conjugate platform, the molecule consists of a heat shock protein 90 (HSP90) binding moiety linked to a phosphoinositide 3-kinase (PI3K) inhibitor payload. This dual-targeting approach leverages the selective overexpression of activated HSP90 in the tumor microenvironment to concentrate the PI3K inhibitor within cancer cells. By targeting both the HSP90 chaperone and the PI3K signaling pathway, the conjugate aims to achieve potent anti-tumor activity in solid tumors while minimizing systemic side effects typically associated with broad PI3K inhibition.
