HspE7 + poly-ICLC is an investigational therapeutic vaccine combination developed for the treatment of precancerous and cancerous lesions caused by human papillomavirus (HPV), particularly HPV type 16. The vaccine consists of a recombinant fusion protein that combines heat shock protein 65 (Hsp65) with the HPV type 16 E7 antigen to stimulate cellular immune responses against infected or transformed cells. Poly-ICLC acts as an adjuvant and is a synthetic double-stranded RNA complex that functions as a Toll-like receptor 3 (TLR3) agonist, enhancing innate and adaptive immune responses. This combination has been studied in clinical trials for indications such as cervical intraepithelial neoplasia (CIN), cervical dysplasia, recurrent respiratory papillomatosis, genital warts, cervical cancer, head and neck cancers, anal intraepithelial neoplasia, and uterine cervical cancer[1][4][5][6]. The addition of poly-ICLC significantly increases the immunogenicity of HspE7 by activating TLR3-mediated pathways[1][4].
