Overview

This is a combination therapy consisting of three agents: **HSPPC‑96** is an autologous, tumor-derived peptide vaccine composed of heat shock protein peptide complex 96, also known as vitespen or gp96. It is manufactured from a patient’s own tumor tissue and contains glycoprotein 96 chaperoning tumor-specific antigenic peptides. The vaccine stimulates both innate and adaptive immune responses by presenting these antigens to the immune system, thereby promoting anti-tumor immunity[1][2][3][4]. **Pembrolizumab** is a humanized monoclonal antibody that targets the programmed cell death protein 1 (PD‑1) receptor on T cells, blocking its interaction with PD-L1/PD-L2 ligands and thus enhancing T-cell mediated anti-tumor activity. **Temozolomide** is an oral alkylating agent that induces DNA damage in cancer cells, leading to apoptosis. It is commonly used in glioblastoma multiforme and other brain tumors[6][9]. The combination aims to leverage the immunogenicity of the personalized vaccine with checkpoint inhibition by pembrolizumab and cytotoxic effects from temozolomide. This approach seeks synergistic enhancement of anti-tumor immune responses while directly targeting tumor cells.
