HST-2011 is a first-in-class small molecule allosteric inhibitor of Interferon Regulatory Factor 5 (IRF5), developed by HotSpot Therapeutics using its proprietary Smart Allostery™ platform. IRF5 is a transcription factor that serves as a master regulator of the innate immune response, controlling the expression of type I interferons and other pro-inflammatory cytokines. Historically, IRF5 was considered undruggable due to the lack of a traditional active site; however, HST-2011 targets a naturally occurring allosteric "hotspot" to potently and selectively inhibit its activity. The drug is currently in IND-enabling studies for the treatment of various autoimmune and inflammatory conditions, including systemic lupus erythematosus (SLE), Sjögren's syndrome, and systemic sclerosis.
